Recent advances in the development of recombinant probes has allowed intracellular free Ca21 concentrations to be examined not just at the level of a single cell, but within subcellular compartments and organelles. This is deepening our insight into the cross-talk of organellar and cytoplasmic Ca21 signalling, and unraveling its importance both for the control of organelle function and in generating the spatio-temporal complexity of the cytoplasmic Ca21 changes. In this review, we will focus on three subcellular compartments—the mitochondria, the nucleus and the secretory organelles—on which a wide body of new experimental evidence has been obtained in the past few years.
