HEp-2 cell cultures were treated with media of high osmolarity (HO) and infected with different strains and mutants of Herpes Simplex virus. Post- treatment with HO media altered the plaques morphology and reduced the number of plaques and the viral yield, with a differential effect on viral strains and mutants. Studies on the effect of HO medium on infectious centers formation suggested that both cells, i.e. recruiters and recruitees in the viral plaques, are susceptible to HO treatment.
