Tandem integrations of complete viral genomes, which are not usually found in hamster cells transformed by BK virus (BKV) or circular BKV DNA can be established when hamster cells are transfected with linear BKV DNA with cohesive ends. Treatment of linear BKV DNA with S1 nuclease or use of linear BKV DNA with staggered but non-complementary ends prevents tandem integration. These results support a model in which formation of tandems of free viral DNA is a preliminary step for tandem integration. © 1982.
