Hydrophilic interaction liquid chromatography (HILIC) has become in the last years a leading technique for the separation of small and large polar molecules. This chapter begins with a discussion about the most relevant characteristics of HILIC mode that makes it a complementary and, in some cases, alternative technique to the more traditional reversed- and normal-phase modes. Subsequently, the fundamentals of retention mechanisms in HILIC are revised in light of the most recent findings in the field. The chapter ends with an overview of the materials used as stationary and mobile phases in HILIC and of the applications of this analytical technique in different branches of science.
